RESUMO

The influence of pharmaceutical residues and heavy metals on living organisms has received global attention. The present study assessed the interactive effect of antibiotic residues and heavy metals in soil, as contaminated food with cadmium (Cd) and oxytetracycline (OTC) on the isopod Porcellio leavis. It was fed on fresh plant leaves contaminated with different concentrations of cadmium, Cd+OTC1000 ppm, Cd+OTC2000 ppm and Cd+OTC3000 ppm for 4 weeks. The changes in the feeding patterns, protein, lipid peroxidation (LPO), catalase activity (CAT), and total free amino acids (TFAA) were recorded. There were significant differences in the obtained results where Cd reduced the egestion ratio (ER) however, OTC enhanced this ratio. Biochemical analysis illustrated that combination between OTC and Cd inhibits the toxic effects of Cd at low concentration (1000 ppm), while at high concentration (3000 ppm) raise the toxicity. Detailed studies are required for further understanding of the interaction between OTC and heavy metals, and also its impact on soil animals and for improving soil risk evaluation.


A influência de resíduos farmacêuticos e metais pesados em organismos vivos tem recebido atenção global. O presente estudo avaliou o efeito de resíduos de antibióticos e metais pesados no solo, como alimentos contaminados com cádmio (Cd) e oxitetraciclina (OTC), sobre o isópode Porcellio laevis, o qual foi alimentado com folhas frescas de plantas contaminadas com diferentes concentrações de cádmio, Cd + OTC1000 ppm, Cd + OTC2000 ppm e Cd + OTC3000 ppm, por quatro semanas. As mudanças nos padrões de alimentação, proteína, peroxidação lipídica (LPO), atividade da catalase (CAT) e aminoácidos livres totais (TFAA) foram registradas. Houve diferenças significativas nos resultados obtidos, em que o Cd reduziu a taxa de excreção (ER), no entanto o OTC aumentou essa proporção. A análise bioquímica mostrou que a combinação entre OTC e Cd inibe os efeitos tóxicos do Cd em baixa concentração (1.000 ppm), enquanto, em alta concentração (3.000 ppm), aumenta a toxicidade. Estudos detalhados são necessários para uma maior compreensão da interação entre OTC e metais pesados, e seu impacto sobre os animais do solo, bem como para melhorar a avaliação de risco do solo.

α-glucosidase (EC: 3.2.1.20) from Penicillium chrysogenum Thom ATCC 10106 was induced by GSH at the lower concentrations. H2O2 was inhibitor at all tested concentrations and the IC50 was 92.2%v/v. AMP, ADP and ATP enhanced the activity revealing that α-glucosidase is endothermic enzyme. The chelating agents are ethylenediaminetetraacetate (EDTA), α-α-dipyridyl and o-phenanthroline inhibited the enzyme. IC50 for these three compounds were 7.1, 10.2 and 10.9 mM, respectively. The highest activity of α-glucosidase was recorded at 150 mM phosphate buffer. Mannitol as polyol protected the enzyme against heat inactivation. The five sugars trehalose, lactose, raffinose, glucose and sucrose protected α-glucosidase against thermo-inactivation at 60ºC. Also, sarcosine as a product of glycine provided α-glucosidase with appreciable thermostability at 60ºC.

Pre-clinical data have shown that tissue level effects stemming from bisphosphonateinduced suppression of bone remodeling can result in bone that is stronger yet more brittle. Raloxifene has been shown to reduce bone brittleness through non-cellular mechanisms. The goal of this work was to test the hypothesis that raloxifene can reverse the bone brittleness resulting from bisphosphonate treatment. Dog and mouse bone from multiple bisphosphonate dosing experiments were soaked in raloxifene and then assessed for mechanical properties. Mice treated with zoledronate in vivo had lower post-yield mechanical properties compared to controls. Raloxifene soaking had significant positive effects on select mechanical properties of bones from both vehicle and zoledronate treated mice. Although the effects were blunted in zoledronate bones relative to vehicle, the soaking was sufficient to normalize properties to control levels. Additional studies showed that raloxifene-soaked bones had a significant positive effect on cycles to failure (+114%) compared to control-soaked mouse bone. Finally, raloxifene soaking significantly improved select properties of ribs from dogs treated for 3 years with alendronate. These data show that ex vivo soaking in raloxifene can act through non-cellular mechanisms to enhance mechanical properties of bone previously treated with bisphosphonate. We also document that the positive effects of raloxifene soaking extend to enhancing fatigue properties of bone. (AU)


Los datos preclínicos han demostrado que los efectos a nivel de tejido que se derivan de la supresión del remodelado óseo inducida por bifosfonatos puede dar como resultado un hueso que es más fuerte pero más frágil. Está comprobado que el raloxifeno reduce la fragilidad ósea a través de mecanismos no celulares. El objetivo de este trabajo fue probar la hipótesis de que el raloxifeno puede revertir la fragilidad ósea resultante del tratamiento con bifosfonatos. Se emplearon huesos de perro y ratón de múltiples experimentos con diferentes dosis de bifosfonatos los cuales fueron sumergidos en raloxifeno y luego se evaluaron sus propiedades mecánicas. Ratones tratados con zoledronato in vivo mostraron propiedades mecánicas post-rendimiento más bajas en comparación con los controles. Luego de sumergirlos en raloxifeno se observaron efectos positivos significativos en algunas propiedades biomecánicas tanto en los huesos de ratones tratados con vehículo como con zoledronato. Aunque los efectos se atenuaron en los huesos tratados con zoledronato en relación con los tratados con vehículo, el raloxifeno fue suficiente para normalizar las propiedades a niveles basales. Estudios adicionales mostraron que los huesos sumergidos en raloxifeno tuvieron un efecto positivo significativo en los ciclos de fractura (+ 114%) en comparación con los huesos de ratón sumergido en vehículo. Finalmente, el raloxifeno mejoró significativamente las propiedades de costillas de perros tratados durante 3 años con alendronato. Estos datos muestran que la inclusión ex vivo en raloxifeno puede actuar a través de mecanismos no celulares para mejorar las propiedades mecánicas de huesos previamente tratado con bifosfonatos. También documentamos que los efectos positivos del raloxifeno mejoran las propiedades de fatiga del hueso. (AU)

RESUMO

SOS-Page electrophoresis identifies the various protein fractions under different conditions. This property of the process was utilized to see the pattern of protein fractions among patients suffering from thyroid disease. A group of 1170 subject, comprising of nearly equal proportions of euthyroid, hypothyroid and hyperthyroid individuals, was studied. Most of the subjects were out-patients of Shaikh Zayed Hospital, Lahore. Random blood samples were taken from the subjects usually in the morning between 0800 - 1200 hours and after processing, the serum was stored at -80[C]. These aliquots were used for thyroid profile, bio-chemical analysis and SDS-PAGE electrophoreses. The serum samples of healthy / euthyroid samples showed 13 major protein fractions ranging in molecular weight of 321 to 28 kDa whereas 12 major protein fractions were detected in patients suffering from thyroid diseases. The fractions were found to be enhanced due to increased capillary permeability and increased interstitial fluid. By contrast, markedly reduced fractions were found among hyperthyroids probably due to the factors of negative nitrogen balance, loss of weight, muscle wasting, weaknesses mild hypo-albuminemia and increased anabolism of protein. The findings presented in this study suggest that SDS-PAGE may be a useful tool in the diagnosis of thyroid disorders
